Instructions

This assay requires pre-scheduling with the Flow Cytometry Laboratory. Contact 248-551-8045 (extension 18045) to schedule testing.

Specimen Collection Criteria

Collect: Five 5 mL Green-top Sodium Heparin tubes on the DONOR and one Gold-top SST tube on the RECIPIENT.

Physician Office/Draw Specimen Preparation

Maintain specimens at room temperature (20-26°C or 68-78.8°F) and transport to the Laboratory immediately. Do not store specimens.

Preparation for Courier Transport

Transport: Heparin and SST tubes, at room temperature (20-26°C or 68-78.8°F).

Rejection Criteria

Any Green-top tubes sent on ice.

In-Lab Processing

Specimens should be analyzed immediately.

Storage

Specimen Stability for Testing:

Room Temperature (20-26°C or 68-78.8°F): Specimens are tested immediately upon receipt.
Refrigerated (2-8°C or 36-46°F): Unacceptable
Frozen (-20°C/-4°F or below): Unacceptable

Specimen Storage in Department Prior to Disposal:

Refrigerated (2-8°C or 36-46°F): 7 days

Transplant
T-cell IgG: Less than 43.
B-cell IgG: Less than 57.

Test Methodology

Immunophenotyping by Multiparameter Flow Cytometry.

Interpretation

FCXM-Transplant: includes T-cell IgG in mean channel fluorescence intensity and B-cell IgG in mean channel fluorescence intensity.

Clinical Utility

This procedure can be used to identify IgG anti-lymphocyte antibodies for organ transplantation patients.
