Instructions

This test requires the Cytomegalovirus (CMV) DNA Quantitation by PCR to have a result of at least 1500 copies/mL.

Specimen Collection Criteria

Collect: Two Lavender-top EDTA tubes.

Physician Office/Draw Specimen Preparation

Centrifuge to separate plasma from cells within 8 hours of collection. Transfer plasma to a plastic transport tube and freeze (-20°C/-4°F or below).

Preparation for Courier Transport

Transport: 4.0 mL serum or plasma, frozen (-20°C/-4°F or below). (Minimum: 1.5 mL)

Rejection Criteria

Specimens not collected and processed as indicated.

In-Lab Processing

Test Methodology

Polymerase Chain Reaction/Sequencing.

Interpretation

Codons 457-630 of the UL97 gene and codons 393-1000 of the UL54 gene are sequenced. Mutations associated to ganciclovir, cidofovir, and foscarnet are reported. Mutations in sub-populations below 20 percent of total may not be detected.

NOTE: This test may be unsuccessful if the plasma CMV DNA viral load is less than 1,500 CMV DNA copies per mL of plasma.
